In pediatric patients with low-grade gliomas showing prolonged response or stable disease on tovorafenib, how would you adjust the duration of therapy?

In a patient with responding/stable disease on tovorafenib, what factors would lead you to consider treatment discontinuation early?

• Persistent rash not otherwise meeting labeled criteria for dose reduction/hold
• Persistent organ dysfunction (renal, hepatic) not otherwise meeting labeled criteria for dose reduction/hold
• Growth velocity delay
• Psychosocial impact of prolonged treatment
• Achievement of a durable complete response
• I would not discontinue therapy prior to 26 cycles unless mandated by significant adverse events
• I would continue therapy indefinitely as long as the patient is tolerating it well
• I am not planning to use tovorafenib at this time
